    Carbohydrate and caffeine improves high intensity running of elite rugby league interchange players during simulated match play.

    The study examined the effects of carbohydrate and caffeine ingestion on simulated rugby league interchange performance. Eight male elite rugby league forwards completed two trials of a rugby league simulation protocol for interchange players seven days apart in a randomized crossover design, ingesting either carbohydrate (CHO; 40 g·h-1) or carbohydrate and caffeine (CHO-C) (40 g·h-1 + 3 mg·kg-1) drink. Movement characteristics, heart rate, ratings of perceived exertion (RPE), and countermovement jump height (CMJ) were measured during the protocol. CHO-C resulted in likely to very likely higher mean running speeds (ES 0.43 to 0.75), distance in high intensity running (ES 0.41 to 0.64) and mean sprint speeds (ES 0.39 to 1.04) compared to CHO. Heart rate was possibly to very likely higher (ES 0.32 to 0.74) and RPE was likely to very likely lower (ES -0.53 to 0.86) with CHO-C. There was a likely trivial to possibly higher CMJ in CHO-C compared to CHO (ES 0.07 to 0.25). The co-ingestion of carbohydrate with caffeine has an ergogenic effect to reduce the sense of effort and increase high intensity running capability that might be employed to enhance interchange running performance in elite rugby league players

    Microparticles and Exercise in Clinical Populations

    open access journalMicroparticles (MPs) are shed membrane vesicles released from a variety of cell types in response to cellular activation or apoptosis. They are elevated in a wide variety of disease states and have been previously measured to assess both disease activity and severity. However, recent research suggests that they also possess bioeffector functions, including but not limited to promoting coagulation and thrombosis, inducing endothelial dysfunction, increasing pro-inflammatory cytokine release and driving angiogenesis, thereby increasing cardiovascular risk. Current evidence suggests that exercise may reduce both the number and pathophysiological potential of circulating MPs, making them an attractive therapeutic target. However, the existing body of literature is largely comprised of in vitro or animal studies and thus drawing meaningful conclusions with regards to health and disease remains difficult. In this review, we highlight the role of microparticles in disease, comment on the use of exercise and dietary manipulation as a therapeutic strategy, and suggest future research directions that would serve to address some of the limitations present in the research to dat

    Effects of familiarization on reliability of muscle-activation and gross efficiency in adolescents and adults

    We investigated the effects of familiarization on the reliability of gross efficiency (GE) and lower-limb electromyography (EMG-rms) of adults and adolescents. We also evaluated the relationship between inter-test differences in GE and EMG-rms. Nine adult and nine adolescent cyclists performed three 10-min cycling tests at 50% of peak power output, separated by 48-h. 45-min familiarization visits were performed 24-h after each test. No differences were found across the tests for adult cyclists' GE or EMG-rms, with Coefficient of Variation (CV%) ranging from 2.6% - 2.9% (GE) and 4.3% - 7.4%. Among adolescents, there was an increase in GE between tests 1 and 2 (P 0.05) between the inter-test differences of GE and EMG-rms. GE can be reliably determined; however, adolescents require two exposures to cycling. Without familiarization, adolescent EMG-rms is more variable than adults and would require larger samples to establish differences. The weak relationships between inter-test EMG-rms and inter-test GE questions the link between muscle activation and GE changes

    Glutamine supplementation reduces markers of intestinal permeability during running in the heat in a dose-dependent manner.

    PURPOSE: To examine the dose-response effects of acute glutamine supplementation on markers of gastrointestinal (GI) permeability, damage and, secondary, subjective symptoms of GI discomfort in response to running in the heat. METHODS: Ten recreationally active males completed a total of four exercise trials; a placebo trial and three glutamine trials at 0.25, 0.5 and 0.9 g kg(-1) of fat-free mass (FFM) consumed 2 h before exercise. Each exercise trial consisted of a 60-min treadmill run at 70% of [Formula: see text] in an environmental chamber set at 30 °C. GI permeability was measured using ratio of lactulose to rhamnose (L:R) in serum. Plasma glutamine and intestinal fatty acid binding protein (I-FABP) concentrations were determined pre and post exercise. Subjective GI symptoms were assessed 45 min and 24 h post-exercise. RESULTS: Relative to placebo, L:R was likely lower following 0.25 g kg(-1) (mean difference: - 0.023; ± 0.021) and 0.5 g kg(-1) (- 0.019; ± 0.019) and very likely following 0.9 g kg(- 1) (- 0.034; ± 0.024). GI symptoms were typically low and there was no effect of supplementation. DISCUSSION: Acute oral glutamine consumption attenuates GI permeability relative to placebo even at lower doses of 0.25 g kg(-1), although larger doses may be more effective. It remains unclear if this will lead to reductions in GI symptoms. Athletes competing in the heat may, therefore, benefit from acute glutamine supplementation prior to exercise in order to maintain gastrointestinal integrity

    The physiological, perceptual and neuromuscular responses of team sport athletes to a running and cycling high intensity interval training session

    Purpose: The acute physiological, perceptual and neuromuscular responses to volume-matched running and cycling high intensity interval training (HIIT) were studied in team sport athletes. Methods: In a randomized cross-over design, 11 male team sport players completed 3 × 6 min (with 5 min between sets) repeated efforts of 15 s exercising at 120% speed (sV ˙ O2max) or power (pV ˙ O2max) at V ˙ O2max followed by 15 s passive recovery on a treadmill or cycle ergometer, respectively. Results: Absolute mean V ˙ O2 (ES [95% CI] = 1.46 [0.47–2.34], p 90% V ˙ O2max during the HIIT was higher for running compared to cycling (ES [95% CI] = 1.21 [0.26–2.07], p = 0.015). Overall differential RPE (dRPE) (ES [95% CI] = 0.55 [− 0.32–1.38], p = 0.094) and legs dRPE (ES [95% CI] = − 0.65 [− 1.48–0.23], p = 0.111) were similar, whereas breathing dRPE (ES [95% CI] = 1.01 [0.08–1.85], p = 0.012) was higher for running. Maximal isometric knee extension force was unchanged after running (ES [95% CI] = − 0.04 [− 0.80–0.8], p = 0.726) compared to a moderate reduction after cycling (ES [95% CI] = − 1.17 [− 2.02–0.22], p = 0.001). Conclusion: Cycling HIIT in team sport athletes is unlikely to meet the requirements for improving run-specific metabolic adaptation but might offer a greater lower limb neuromuscular load

    Early Initiation of Antiretroviral Therapy Preserves the Metabolic Function of CD4+ T Cells in Subtype C Human Immunodeficiency Virus 1 Infection

    Background: Immune dysfunction often persists in people living with human immunodeficiency virus (HIV) who are on antiretroviral therapy (ART), clinically manifesting as HIV-1-associated comorbid conditions. Early ART initiation may reduce incidence of HIV-1–associated immune dysfunction and comorbid conditions. Immunometabolism is a critical determinant of functional immunity. We investigated the effect of HIV-1 infection and timing of ART initiation on CD4+ T cell metabolism and function. // Methods: Longitudinal blood samples from people living with HIV who initiated ART during hyperacute HIV-1 infection (HHI; before peak viremia) or chronic HIV-1 infection (CHI) were assessed for the metabolic and immune functions of CD4+ T cells. Metabolite uptake and mitochondrial mass were measured using fluorescent analogues and MitoTracker Green accumulation, respectively, and were correlated with CD4+ T cell effector functions. // Results: Initiation of ART during HHI prevented dysregulation of glucose uptake by CD4+ T cells, but glucose uptake was reduced before and after ART initiation in CHI. Glucose uptake positively correlated with interleukin-2 and tumor necrosis factor-α production by CD4+ T cells. CHI was associated with elevated mitochondrial mass in effector memory CD4+ T cells that persisted after ART and correlated with PD-1 expression. // Conclusions: ART initiation in HHI largely prevented metabolic impairment of CD4+ T cells. ART initiation in CHI was associated with persistently dysregulated immunometabolism of CD4+ T cells, which was associated with impaired cellular functions and exhaustion

    Image reconstruction of oxidized cerebral cytochrome C oxidase changes from broadband near-infrared spectroscopy data

    In diffuse optical tomography (DOT), overlapping and multidistance measurements are required to reconstruct depth-resolved images of oxy- ([Formula: see text]) and deoxy- (HHb) hemoglobin concentration changes occurring in the brain. These can be considered an indirect measure of brain activity, under the assumption of intact neurovascular coupling. Broadband systems also allow changes in the redox state of cytochrome c oxidase (oxCCO) to be measured, which can be an important biomarker when neurovascular coupling is impaired. We used DOT to reconstruct images of [Formula: see text], [Formula: see text], and [Formula: see text] from data acquired with a broadband system. Four healthy volunteers were measured while performing a visual stimulation task (4-Hz inverting checkerboard). The broadband system was configured to allow multidistance and overlapping measurements of the participants' visual cortex with 32 channels. A multispectral approach was employed to reconstruct changes in concentration of the three chromophores during the visual stimulation. A clear and focused activation was reconstructed in the left occipital cortex of all participants. The difference between the residuals of the three-chromophore model and of the two-chromophore model (recovering only [Formula: see text] and [Formula: see text]) exhibits a spectrum similar to that of oxCCO. These results form a basis for further studies aimed to further optimize image reconstruction of [Formula: see text]
